Interestingly, Genistein significantly reduced radiation-induced oxidative damage in normal tissue cells (human embryonic lung fibroblasts MRC-5 cells), exerting a radiation protective effect on normal tissue cells.
When further exploring its mechanism, it was found that Genistein is structurally similar to DNA methyltransferase 1 and can competitive inhibit the methylation of the Keap1 promoter region in A549 cells, thereby increasing the expression level of Keap1, weakening the Keap1/Nrf2 signaling pathway, and promoting oxidative damage. However, the methylation base level of the Keap1 promoter in normal tissue cells is relatively low, and Genistein will not have a significant impact on it.
In addition, Genistein can also promote the nuclear translocation of Nrf2, exerting a radiation protection effect. These findings have revealed the molecular mechanism of Genistein's selective radiation sensitization effect, providing new methods and strategies for improving the efficacy of tumor radiotherapy while protecting normal tissues.
