What is Desloratadine?
Desloratadine is an active metabolite of loratadine obtained by decarboxylation in vivo. It is a new third-generation antihistamine with strong antiallergic effect, rapid onset and lasting effect. It has affinity and selectivity for peripheral H1 receptor, and is a new product for treating seasonal allergic rhinitis and chronic urticaria. In vitro model experiments of human H1 receptor expression confirmed that its antagonistic activity was 50 ~ 200 times that of loratadine, fexofenadine or cetirizine, and it was safe and effective, well tolerated by patients, with a half-life of 21 ~ 24h. Compared with terfenadine and Astemizole (both of which were banned from commercialization in 1999), desloratadine did not produce any effects related to QT interval and ventricular rate.
Desloratadine powder is a white-like crystalline powder, odorless and bitter. Not easy to absorb moisture. The melting point is 150 ~ 151℃. Soluble in chloroform, methanol and ethanol, soluble in dichloromethane, ethyl acetate and acetone, almost insoluble in water.
Desloratadine belongs to the tricyclic long-acting third generation histamine Antagonist, which selectively acts on H1 receptor. Receptor binding data show that if the concentration is 2-3ng/ml(7nmol), it will have a meaningful interaction with human histamine H1 receptor. What is the pharmacological action of Desloratadine?
Desloratadine is a non-sedating, long-acting tricyclic antihistamine and an active metabolite of Loratadine. It can relieve symptoms related to allergic rhinitis or chronic idiopathic urticaria by selectively antagonizing peripheral H1 receptors. In addition, in vitro studies have shown that Desloratadine can inhibit the release of histamine from human mast cells. Animal studies have shown that Desloratadine is not easy to pass through the blood-brain barrier.
