Allantoin powder belongs to imidazole heterocyclic compounds and is a derivative of uric acid. It is a non-toxic, tasteless, non-irritating, non-allergenic white crystal. It crystallizes in water as a single prism or colorless crystalline powder. Melting point: 226-240℃. It is soluble in hot water, hot alcohol and dilute sodium hydroxide solution. It is slightly soluble in water and alcohol at room temperature, and difficult to dissolve in organic solvents such as ether and chloroform; its saturated aqueous solution (concentration of 0.6%) is slightly acidic; the pH is 5.5. It is stable in aqueous solutions with a pH value of 4-9. It is also stable in non-aqueous solvents and dry air; it can be decomposed by boiling in a strong alkaline solution and under sunlight exposure.

Allantoin exists in uterine fluid, fetal urine and some plants. However, the cost of extracting allantoin from these substances is too high. The current methods for synthesizing allantoin are: potassium permanganate oxidation of uric acid method; heating dichloroacetic acid and urea to synthesize allantoin; direct condensation of glyoxylic acid and urea.
Allantoin was extracted from the preparations using distilled water containing 70% (v/v) methanol and separated from interfering substances by reverse phase chromatography. Separation was performed using an amino column (250 × 4.5 mm i.d.) and a mobile phase of distilled water containing 70% (v/v) acetonitrile. Quantification was performed using a UV detector at 220 nm. The relative standard deviation of the assay was approximately 1.7% (n = 10) and the average recovery of laboratory prepared samples was 100%.
