Due to the medicinal value of retinaldehyde, the demand for microbial synthesis of bio-based retinaldehyde is increasing. However, in the current research on the synthesis of vitamin A by microbial methods, the obtained products contain a mixture of retinaldehyde, retinol and retinoic acid. The structures and properties of the products are relatively similar, which increases the cost of downstream separation and purification.

Saccharomyces cerevisiae contains over 30 kinds of oxidoreductases. The accumulation of retinaldehyde in Saccharomyces cerevisiae is achieved through the combined inactivation of alcohol dehydrogenases (Adh6, Adh7, Sfa1 and Gre2) and an aldehyde dehydrogenase (Hfd1). The mevalnate (MVA) pathway was optimized by using a combined method. On the basis of the yeast chassis cells, CrtE/YB/I from Rhodochroma was heterologous expressed, improving the synthesis of β -carotene in yeast. In addition, the BCMO from the Marine bacterium 66A03 was further integrated. By integrating the ATP-dependent citrate lyase ACL from the esterolytic yeast, citric acid was converted into acetyl-CoA in the cytoplasm of the cell, thereby obtaining an engineered strain of Saccharomyces cerevisiae with high retinaldehyde yield. 69.13±5.82 mg L−1 retinaldehyde (>99% purity) was obtained in the shake flask.
