SLU-PP-332 (SLU-PP) enhances mitochondrial function and cellular respiration, increases type IIa oxidative skeletal muscle fibers, and enhances exercise tolerance in skeletal muscle cell lines. SLU-PP-332 has the potential to be used in the study of metabolic diseases and the improvement of muscle function, and can be used to improve metabolic disorders and aging.
SLU-PP-332 is a non-toxic pan ERRα/β/γ (estrogen receptor-related receptors α/β/γ) agonist with significant ERRα activity (EC50 = 98, 230 & 430 nM against ERRα, ERRβ & ERγ) and acts as an exercise mimetic. SLU-PP-332 displays selectivity over ERα, ERβ and several other closely related nuclear receptors. It induces Ddit4 expression and enhances mitochondrial respiration in C2C12 myocytes (10 µM) and exercise endurance in mice (50 mg/kg bid, ip) in an ERRα-dependent manner.
SLU-PP-332 is a potent and selective pan estrogen related receptor (ERR) agonist (EC50 values are 98 nM, 230 nM and 430 nM for ERRα, ERRβ and ERRγ respectively). In a mouse model of metabolic syndrome SLU-PP-332 increases energy expenditure and fatty acid oxidation, decreases fat mass, and improves insulin sensitivity. Also increases mitochondrial respiration in vitro.
