Flumazenil powder is the first antagonist of benzodiazepines. It is used to counteract the sedative effects and disorientation caused by overdose of benzodiazepines. It also has anticonvulsant and antiepileptic effects. It can also be used in the recovery period after halothane anesthesia and in encephalopathy of cirrhosis caused by alcohol poisoning. It is used as a diagnostic drug for unexplained loss of consciousness and to differentiate between benzodiazepines and other drug poisoning or brain damage.
A HPLC method for the determination of flumazenil was established. Methods: An ODS reverse phase column was used, the mobile phase was methanol-water (50∶50), the flow rate was 1.0 mL·min-1, vanillin was used as the internal standard, and the detection was performed by an ultraviolet detector (λ=243 nm). Results: The linear relationship was good in the range of 80-400 ng (r=0.999 9), and the average recovery was 98.6%. Conclusion: The RP-HPLC method is simple, rapid, accurate, and suitable for the determination of flumazenil.
Flumazenil was synthesized by a new route: under standard conditions of nucleophilic fluorination, 19F isotope exchange was carried out with the complex [K/K2.2.2]+18F-. The synthesis conditions were: solvent dimethyl sulfoxide, reaction time 5 min, temperature 130°C, flumazenil sample weight 2 mg, molar ratio of potassium to water 1:1, reaction time 1 h, and fluorination efficiency of 628% (n=8).
