How to produce Cidofovir API raw material?
Compound (I) was dissolved in 10ml of Triethyl phosphate, and 0.40ml of C1CH2P (O) Cl2 was added while stirring. After addition, the mixture was stirred for 16 hours. 80mL of substance A was added, and the precipitate was collected by filtration. The precipitate was washed with substance A and dried under vacuum at 13Pa. Then dissolve in 20ml of water and reflux for 8 hours. Neutralize with Triethylamine and evaporate the solvent under vacuum at 2kPa. The residue is dissolved in 1.5ml of water. Take 0.3ml of this solution and perform column chromatography on silica gel (such as SeparonSIXCl8, 7 μ) with acylation. Use a 0.05mol/L buffer solution of triethylamine bicarbonate with a pH of 7.5 as the eluent, and wash until the salt is completely removed. Then use the buffer solution containing 10% (V/V) Methanol as the eluent, with a flow rate of 2ml/min. Combine the eluents and evaporate the solvent under reduced pressure. Add 10ml of 2ml/L sodium hydroxide solution and heat at 80 ℃. Neutralize with H+type cation exchange resin. Filter and evaporate the filtrate under a vacuum of 2Pa to remove the solvent. The residue was separated by column chromatography to obtain Cidofovir API raw material with a yield of 50% to 60%.
