What is Caspofungin?

Caspofungin powder  is the first echinocandin product approved for the treatment of invasive fungal infections. Caspofungin is a semi-synthetic lipopeptide (echinocandin) compound derived from the fermentation products of Glarea Lozoyensis. Caspofungin acetate inhibits the synthesis of β(1,3)-D-glucan, a fundamental component of the cell walls of many filamentous fungi and yeasts. β(1,3)-D-glucan is absent in mammalian cells. In vitro pharmacological studies have shown that Aids-058650 has antibacterial activity against many pathogenic Aspergillus and Candida species. In vitro and in vivo pharmacological experiments have verified that Caspofungin possesses excellent antibacterial activity against common opportunistic pathogens such as Candida and Aspergillus. Its mechanism of action is to inhibit the biosynthesis of 1,3-β-glucan in the fungal cell wall and disrupt the structure of the fungal cell wall; clinical applications have also confirmed that this component has significant therapeutic effects on various types of candidiasis and aspergillosis.

The emergence of resistance in fungal pathogens to traditional antifungal products has led to a growing need for novel therapeutics. Caspofungin powder is a prime example of such a novel compound, altering cell wall biosynthesis by inhibiting the synthesis of β-1,3-D-glucan. While significant progress has been made in understanding the mechanism of action of Aids-058650, its effects on the biophysical properties of fungal cells remain poorly understood. This study uses atomic force microscopy (AFM) to demonstrate that Cancidas (tm) significantly alters the cell surface properties of "Candida albicans". CASPOFUNGIN induces significant changes in cell morphology and structure, which are closely related to a decrease in cell wall mechanical strength. Furthermore, we found that this product induces a large exposure of the cell adhesion protein Als1 on the cell surface, leading to increased cell surface hydrophobicity-both features that trigger cell aggregation. This behavior was not observed in yeast strains lacking Als1, suggesting that this protein plays a crucial role in determining the aggregation phenotype of "Candida albicans".

 

 

How Is Caspofungin Metabolized In The Liver?

Cancidas (tm) exhibits bactericidal activity against Candida spp. (including triazole-resistant strains) and bacteriostatic activity against Aspergillus spp. The pharmacokinetics of Caspofungin are triphasic and nonlinear. Subsequently, the product is gradually released from extravascular tissues and slowly metabolized in the liver. The product is hydrolyzed in the liver and slowly metabolized by N-acetylation into two inactive metabolites; dose reduction is recommended in cases of severe hepatic impairment. Aids058650 also spontaneously degrades into open-ring compounds.

Like other echinocandins, Caspofungin powder exerts its effects by noncompetitively inhibiting β-(1,3)-D-glucan synthase, a key component in fungal cell wall synthesis; this enzyme is absent in mammalian cells.
