We used comparative molecular modeling and ligand docking methods to analyze the three-dimensional structures of quinidine and chloroquine in the intracellular domain of Kir2.1. The simulation results predicted that chloroquine effectively blocks potassium flow by binding to the center of the Kir2.1 ion-permeable vestibule. In contrast, quinidine binds to the vestibule side and only partially blocks ion movement.
