What are the relevant studies on Cinacalcet Hcl powder?
In vitro
Cinacalcet HCl powder represents a new type of compound used to treat hyperthyroidism, which is a calcium mimetic agent that reduces the synthesis and secretion of parathyroid hormone (PTH) by increasing the sensitivity of parathyroid calcium sensitive receptors (CaR) to extracellular calcium ions. Cinacalcet HCl, as a therapeutic agent for secondary hyperparathyroidism, has potential advantages because it simulates the action of extracellular calcium ions to inhibit PTH secretion, and even in cases of hyperphosphatemia, there is no risk of causing hypercalcemia and/or hyperphosphatemia. CinacalcetHCl induces a concentration dependent increase in calcium ions in the cytoplasm of human embryonic kidney cells expressing CaSR. In bovine parathyroid cells and a buffer containing 0.5 mM calcium and AMG073 (3nM – 1 μ M), PTH levels were reduced concentration dependently, with an IC50 of 27 nM.
In vivo
oral administration of Cinacalcet HCl powder at doses of 1, 3, 10, and 30 mg/kg in 20% sulfobutyl ether β - cyclodextrin sodium in normal rats resulted in a significant dose-dependent decrease in PTH levels after 1 to 4 hours of administration. At 8 hours, compared with the control group, Cinacalcet HCl at doses of 10 and 30 mg/kg significantly reduced PTH levels, and this effect disappeared at 24 hours. Cinacalcet HCl was orally administered at doses of 3, 10, and 30 mg/kg, and significant reductions in serum calcium levels were observed at 4, 8, and 24 hours, respectively. The transient decrease in serum phosphorus levels can only be observed at high doses of Cinacalcet HCl. In addition, in rats, an increase in calcitonin levels was observed at a concentration of 40 mg/kg Cinacalcet HCl, accompanied by inhibition of PTH. In normal rats, rapid dose-dependent reductions in PTH and calcium levels were observed in 5 out of 6 nephrectomized rats after oral administration of Cinacalcet HCl. In addition, compared with the control group, oral administration of 5 and 10 mg/kg Cinacalcet HCl for 4 weeks significantly reduced the weight of parathyroid glands.
