Chlorpheniramine maleate powder is used for colds and skin and mucosal allergic diseases. It mainly treats urticaria, eczema, dermatitis, drug eruption, pruritus, neurodermatitis, insect bites, solar dermatitis, and can also be used for allergic dermatitis, vasomotor rhinitis, drug and food allergies, etc. In addition to having a strong competitive blocking effect on histamine H1 receptors on allergic target cells and playing an anti-allergic role by antagonizing H1 receptors, it also has an anti-M cholinergic receptor effect. Therefore, after taking the medicine, symptoms such as dry mouth, constipation, thickened sputum, and dry nasal mucosa may occur.
Obtained by two-step condensation of 2-p-chlorobenzylpyridine. Add sodium ammonia and anhydrous toluene to a dry reaction pot, heat to reflux, and drop 2-p-chlorobenzylpyridine and bromoacetaldehyde diethyl acetal at the same time. After addition, reflux reaction for 8 hours. Cool and let stand, take the toluene solution, dissolve the solid with a small amount of water and extract with toluene. Combine the extract with the toluene solution, wash with water until neutral, recover toluene, and obtain 3-p-chlorobenzene-3-(2-pyridyl)-propionaldehyde diethyl acetal.
Mix it with DMF and add it to the reaction pot, and add a mixture of formic acid and water under stirring. Heat to 110-120℃ for reaction for 2h, and 150-160℃ for reaction for 3h. Cool, recover DMF under reduced pressure, dilute with hot water, and adjust to pH14 with alkali solution. Then extract with toluene, wash the extract with water until neutral, recover toluene and distill under reduced pressure, collect the 150-190℃ fraction to obtain chlortrimetone.
