Alogliptin powder is a dipeptidyl peptidase-4 (DPP-4) inhibitor that slows down the inactivation of the incretin hormones GLP-1 (glucagon-like polypeptide-1) and GIP (glucose-dependent insulinotropic polypeptide). When blood glucose concentrations are normal or elevated, GLP-1 and GIP can increase the synthesis and release of insulin by pancreatic β cells through an intracellular signaling pathway involving cyclic adenosine monophosphate.
The classical method for the partial reduction of 6 to 7 using Pd/C as a heterogeneous hydrogenation catalyst gave 70-92% yields. The optimization of the partial reduction was investigated to suppress overreduction to racemic nipecoamide. The stirring speed and hydrogen pressure in the reaction mixture were found to be important to obtain 7 in acceptable yields and with constant reproducibility.

CHEMCAT Corporation in MeOH with a hydrogen pressure of 0.1 MPa at 40 °C for 21 h. The product was obtained in 60% yield in two steps on a 70 kg scale of 6,7·TsOH and was directly used as substrate for the next asymmetric hydrogenation reaction.
