5-Fluoroindole production can be carried out through the following steps:
Step 1: Preparation of intermediate crude product
Add 7.2 grams of 5-fluoro-2-nitrotoluene (compound 1b, 0.046 mol) and 27.4 grams of DMFDMA (0.232 mol) to 80 ml of acetonitrile, then raise the reaction temperature to 85 ° C to 95 ° C and reflux for 5 hours. After cooling the reaction, spin dry the solvent, and wash with water and trace amounts of ethanol, 9.3 grams of crude product 2b (0.0443 mol, yield 96.2%) can be obtained. This is done by reacting 5-fluoro-2-nitrotoluene and DMFDMA in a specific solvent and temperature to generate the desired intermediate.
Step 2: Preparation of 5-fluoroindole
Add the intermediate 2b obtained in the first step to 147ml of methanol containing 10wt% hydrazine (hydrazine content 0.45mol, 10eq), then raise the temperature to 50 ℃, react for 8 hours, and then cool to room temperature. Finally, the solvent was spin dried to obtain 5.7 grams of product 3b (0.0424 mol, yield 95.8%). This step involves further reaction of the intermediate with hydrazine to generate the product.
