Lesinurad

The synthesis of lesinurad can generally be obtained by the epoxidation of cyclohexane. Cyclohexane is first reacted with hydrogen peroxide (H2O2). In the presence of an alkaline catalyst, cyclohexane is oxidized to cyclohexane-2, 3-epoxide. The epoxide is then reacted with silver carbonate to obtain the product of lesinurad. The reaction temperature and pH value need to be controlled during the reaction to obtain high yield and purity. The synthesis of lesinurad should be carried out in strict accordance with laboratory operating procedures and ensure safety and environmental friendliness.

 

MECHANISM

The mechanism of action of lesinurad is mainly by inhibiting the function of transporters involved in uric acid reabsorption in the kidney, helping the kidney to excrete uric acid. It is a uric acid transport inhibitor and is primarily used to treat high blood uric acid levels associated with gout. It can selectively inhibit the function of uric acid transporter 1(URAT1) and organic anion transporter (OAT), thereby reducing the reabsorption of uric acid in the kidney and increasing the excretion of uric acid. This can reduce the level of uric acid in the blood, which can relieve the symptoms of gout. However, lesinurad is often used with drugs such as allopurinol or febuxostat to achieve better treatment results.
