6-Diazo-5-oxo-L-nor-Leucine powder is an antibiotic isolated from Streptomyces, a glutaminase antagonist with an IC50 value of about 1 mM for cKGA (kidney-type glutaminase). 6-Diazo-5-oxo-L-norleucine (DON for short) is a glutamine antagonist. Due to its structural similarity to glutamine, 6-Diazo-5-oxo-L-norleucine can enter its catalytic center and inhibit its activity by covalent binding or alkylation. It can very broadly and effectively inhibit the activity of various enzymes with glutamine as a substrate. At present, this compound is being studied for the treatment of diseases such as tumors.

6-Diazo-5-oxo-L-nor-Leucine powder is a glutamine antagonist that inhibits tumor growth. At present, its anti-tumor effect is increasingly valued, and people focus on the preparation of 6-diazo-5-oxo-L-norleucine prodrugs. Only by introducing a targeting group with a specific recognition function, designing and synthesizing tumor-targeted 6-diazo-5-oxo-L-norleucine prodrugs, and selectively delivering drugs to the lesion site, can the toxic effects on normal cells, tissues and organs be reduced, thereby increasing its therapeutic window.
6-Diazo-5-oxo-l-norleucine (DON) is a glutamine antagonist with robust anticancer efficacy; however, its therapeutic potential was hampered by its biodistribution and toxicity to normal tissues, specifically gastrointestinal (GI) tissues. To circumvent DON's toxicity, we synthesized a series of tumor-targeted DON prodrugs designed to circulate inert in plasma and preferentially activate over DON in tumor.
Our best prodrug 6 (isopropyl 2-(6-acetamido-2-(adamantane-1-carboxamido)hexanamido)-6-diazo-5-oxohexanoate) showed stability in plasma, liver, and intestinal homogenates yet was readily cleaved to DON in P493B lymphoma cells, exhibiting a 55-fold enhanced tumor cell-to-plasma ratio versus that of DON and resulting in a dose-dependent inhibition of cell proliferation.
