Pidotimod powder is a representative of chemically synthesized immunostimulants, and its structure is similar to dipeptide. The drug has the advantages of rapid distribution and excretion, no accumulation in the body, good tolerance, and no restrictions on the age of applicable patients.
First, L-cysteine (1) and formaldehyde are subjected to dehydration cyclization to obtain L-thiazolidine-4-carboxylic acid (2). L-pyroglutamic acid (3) and pentachlorophenol are reacted in solvent N,N-dimethylformamide (DMF) under the action of condensation agent dicyclohexylcarbonyldiimide (DCC) to obtain L-pyroglutamic acid pentachlorophenol ester (4). Compound (2) and compound (4) are subjected to dehydration condensation under the action of solvent DMF and acid-binding agent triethylamine to obtain pidotimod.
Pidotimod was successfully separated from the endogenous interference in plasma using equal-volume 10 % perchloric acid as protein precipitation agent, and 0.01 M sodium dihydrogen phosphate (pH 4.0 with concentrated phosphoric acid)-methanol-isopropanol (97:2:1, v/v) as mobile phase. After validation of specificity, linearity, recovery, precision, accuracy and stability, this method has been successfully applied to a pharmacokinetic study in rats.
