N4-Acetylcytosin powder is a white or beige solid at room temperature and pressure. It belongs to the pyrimidine derivatives. Like pyridine, pyrimidine also retains aromaticity and is often used as a pharmaceutical intermediate, a raw material for photosensitizers, etc. Add acetic anhydride (2.1 ml, 22.05 mmol) to a stirred solution of cytosine (500 mg) in pyridine (2.5 ml), stir the resulting mixture overnight at room temperature, dilute the mixture with EtOAc (2.0 mL), stir the mixture again at room temperature for 30 minutes, filter the obtained white solid, wash the solid with EtOAc, and then dry the solid under vacuum to obtain the product N4-acetylcytosine.

N4-Acetylcytosin powder plays an important role in the synthesis of pyrimidine anti-tumor drugs and anti-AIDS drugs. In the synthetic transformation, the nitrogen atom on the pyrimidine ring can react with methyl iodide to obtain a product with methylation of the amide nitrogen atom.
Pyridine (2.2 ml, 27.3 mmol) and DMAP (10 mg) were added to a suspension of cytosine (2.00 g) in toluene (6 ml) at room temperature, acetic anhydride (2.17 g) was diluted with toluene (3.5 ml), and the resulting mixture was slowly added to the mixture. After the addition, the reactants were heated to 50-55 degrees, and the mixture was stirred for 6 hours until the reaction was completed. The reaction mixture was cooled to room temperature, the product was filtered, and the product was washed with toluene (3.5 ml) and water (2 x 3 ml).
Under argon, cytosine (0.4 mmol, 1.0 equiv) and Cs2CO3 (0.8 mmol, 2.0 equiv) were added to a 25 ml Schlenk tube, followed by the addition of DMSO (3 ml) under stirring. MeI (1.2 mmol, 3.0 equiv) was then added, stirred for 24 hours, and the reaction mixture was filtered through silica gel. The combined organic layers were removed by vacuum distillation. The residue was purified by silica gel chromatography (petroleum ether/ethyl acetate = 1:3) to obtain the product.
