What is Metapirone?

Metapyron is a white crystalline powder with a melting point of 50–53°C. It is soluble in ethanol and chloroform, and slightly soluble in water. Metyrapone API is an adrenocortical insufficiency inhibitor, belonging to the 11-β-hydroxylase inhibitor class. It inhibits Corticosteroids production and is used to treat adrenocortical tumors, Cushing's syndrome, and other conditions. It can also be used in hypothalamic-pituitary-adrenal (HPA) examinations.

What is the mechanism of action of Metapyron?

It mainly blocks the final step of Corticosteroids biosynthesis by inhibiting the 11-β-hydroxylase in the adrenal cortex. As a result, CORTEXOLONE is converted into Corticosteroids. This inhibitory effect leads to a decrease in plasma Corticosteroids levels, thereby relieving the negative feedback inhibition of Corticosteroids on the pituitary gland, and stimulating the pituitary gland to secrete more adrenocorticotropic hormone. In the presence of continuous enzyme inhibition, the increase in ACTH will stimulate the adrenal cortex to synthesize and release the precursor substances of corticosteroids - CORTEXOLONE and Desoxycorticosterone. The concentrations of these metabolites in the blood and urine will significantly increase. Quantitative analysis can be performed by measuring urinary 17-hydroxycorticosteroids. Therefore, the Metyrapone API mechanism is used as a diagnostic test for evaluating the pituitary ACTH reserve function.
