Describe	Agmatine sulfate plays a regulatory role on multiple targets, such as neurotransmitter systems, ion channels, and nitric oxide synthesis. It is an endogenous agonist of the imidazoline receptor and an inhibitor of NO synthase.
In vitro studies	Agmatine binds to α2-adrenergic and imidazoline receptors and stimulates the release of catecholamines from adrenal chromaffin cells. Its biosynthetic enzyme, arginine decarboxylase, is present in the brain. Locally synthesized agmatine is an endogenous agonist of imidazoline receptors and a non-catecholamine ligand for α2-adrenergic receptors, acting as a neurotransmitter [1]. Agmatine is synthesized in the brain and stored in synaptic vesicles of region-selective neurons, where it accumulates by uptake, is released by depolarization, and is inactivated by agrochemical enzymes. Agmatine inhibits nitric oxide synthase and induces the release of some peptide hormones [2]. Agmatine 4-(aminobutyl)guanidine is produced by decarboxylation of L-arginine by the enzyme arginine decarboxylase. Agmatine is a competitive inhibitor of all NOS isozymes, but not the NO precursor. Ki values ​​are approximately 660 μM (NOS I), 220 μM (NOS II), and 7.5 mM (NOS III) [3]. Agmatine stimulates nitrite production to be three times higher than the basal nitrite formation by endothelial cells. Agmatine displaces [3H]-deazuridine from the endothelial cell membrane and has been found to induce cytosolic calcium transients in endothelial cells. The transients can be downregulated by repeated exposure to agmatine but are not affected by norepinephrine pretreatment [4].
In vivo studies	Agmatine produced antidepressant-like effects when assessed in the forced swim test and the tail suspension test in mice (dose range 0.01-50 mg/kg, ip) without concomitant changes in locomotion in the open field [5]. In ischemic stroke, agmatine protects the blood-brain barrier, which can be monitored in vivo by quantitative permeability using dynamic contrast-enhanced magnetic resonance imaging [6]. Agmatine greatly enhanced the antidepressant-like effects of MK-801, reinforcing the notion that this compound modulates NMDA receptor activation [7].
Animal experiments	Rats: Thirty-four male Sprague-Dawley rats underwent transient MCA occlusion for 90 minutes. Immediately after reperfusion, agmatine (100 mg/kg) or saline was injected intraperitoneally into the agmatine-treated group (n = 17) or control group. MR imaging was performed after reperfusion [6]. Mice: Mice were pretreated with a series of subeffective doses of fluoxetine (1, 2.5, and 5 mg/kg, po; selective serotonin reuptake inhibitor), imipramine (0.01, 0.05, and 0.1 mg/kg, po; tricyclic). antidepressant), bupropion (0.1, 0.5, and 1 mg/kg, po; a dopamine reuptake inhibitor with subtle activity on noradrenergic reuptake), or MK-801 (0.0001, 0.0005, and 0.001 mg/kg, po; a noncompetitive NMDA receptor antagonist) and immediately thereafter, a subeffective dose of agmatine (0.0001 mg/kg po) or vehicle was given. 60 min later, the animals were subjected to behavioral testing.[7]
