PNC27 peptide (also known as PNC-27) is a chimeric peptide consisting of 32 amino acid residues and two functional domains: amino acids 12-26 from the HDM-2 (MDM2) binding region of the p53 tumor suppressor protein, linked to a membrane-penetrating sequence (initially penetratin, later optimized to the membrane-resident peptide MRP). It was initially designed as a decoy peptide to block the p53-HDM2 interaction in cancer cell nuclei, but it was later discovered that it exerts its anti-cancer effect through a novel membrane-targeting mechanism.
PNC27 kills cancer cells through a membrane-targeting process independent of p53 (called "poptosis," a combination of perforation and necrosis):
Selective binding: Targets the membrane and binds to HDM-2 (overexpressed in cancer cells but absent in normal cells).
Perforation: The PNC27-HDM2 complex oligomerizes and inserts into the membrane to form transmembrane channels.
Membrane disruption: These channels lead to rapid membrane permeability, ion imbalance, and cell lysis (necrosis).
Mitochondrial damage: It can also enter the cell and damage the mitochondrial membrane, amplifying cytotoxicity.
This mechanism differs from traditional chemotherapy and many targeted products because it is independent of p53 function, making it equally effective against cancers with p53 mutations or deficiency.
