It was discovered by Henry W Dion and others in 1956, and it was suggested that it might be used for cancer treatment. This anti-tumor effect has been confirmed in different animal models. DON has been tested as a chemotherapy drug in different clinical studies. In 2019, studies showed that DON can kill tumor cells, reverse disease symptoms and improve the overall survival rate of experimental glioblastoma in mice when combined with a ketogenic diet with calorie restriction.
6-Diazo-5-oxo-L-norleucine (DON) is a glutamine antagonist, which was originally isolated from Streptomyces in Peruvian soil samples. This diazo compound is biosynthesized from lysine by three enzymes in bacteria.DON is a cytotoxic inhibitor of many enzymes of nucleotide synthesis. It could be shown in vitro that DON treatment led to apoptosis, or programmed cell death. Different pathways were investigated; it could be shown that the inner mitochondrial membrane was damaged, and that single strand DNA breaks occurred.

1. 6-Diazo-5-oxo-L-norleucine (Diazooxonorleucine, L-6-Diazo-5-oxonorleucine, DON) is an antibiotic isolated from Streptomyces, A glutaminase antagonist has an IC50 value of about 1 mM for cKGA (renal type glutaminase). 6-Diazo-5-oxo-L-norleucine has analgesic, antibacterial, antiviral and anticancer properties.
