Palmatine hydrochloride is an isoquinoline alkaloid compound, its English name derived from plants of the genus "Stephania" in the palm family. This substance is widely found in medicinal plants such as "Coptis chinensis", and has structural characteristics similar to berberine and purslane, but it has not been found in "Ranunculus spp" .

Palmatine HCL is a typical isoquinoline alkaloid, mainly found in the rhizomes of natural Chinese herbal medicines such as "Phellodendron chinense", "Coptis chinensis", "Smilax china", and "Nandina domestica", and is an effective component in traditional Chinese medicine. It possesses pharmacological effects such as clearing heat and detoxifying, and antibacterial, antiviral, and antifungal effects. Additionally, Palmatine chloride also exhibits certain antitumor activity, strongly inhibiting oxygen uptake by mouse ascites cancer cells.   

Its hydrochloride and hydroiodide derivatives are used in pharmaceutical testing, for example, in the determination of "Coptis chinensis" capsule components using high-performance liquid chromatography (HPLC). Studies show that the recovery rate of palmatine hydrochloride in traditional Chinese medicine extraction can reach over 97%, and it serves as a key indicator in the quality analysis of products such as rotundine. This compound is often detected in conjunction with other components such as total alkaloids from Coptis chinensis, and is also involved in pharmacological studies related to colon and cholinesterase.

Palmatine hydrochloride  is a natural active component derived from traditional Chinese medicine. Firstly, the absorption and fluorescence spectra of PaH were measured using UV-Vis spectrophotometer and RF-1500PC spectrophotometer respectively. The amount of oxygen free radicals (¹O₂) generated by PaH was measured using the DPBF method. The black toxicity of PaH was evaluated using the MTT method. The absorption rate of PaH by HT-29 cells was measured at various time intervals. The location of PaH within HT-29 cells was observed using a confocal laser fluorescence microscope. The photodynamic treatment was performed by exposing HT-29 cells cultured with PaH to visible light (470 nm) emitted from an LED light source. After 24-hour photodynamic treatment, the phototoxicity was evaluated using the MTT method. After 18-hour photodynamic treatment, cell apoptosis was observed using the Alexa Fluor 488/PI double staining cell analysis. The results showed that PaH exhibited absorption peaks in the 400-500 nm visible light region and fluorescence peaks at 406 nm in the 365 nm range. PaH was activated by the 470 nm visible light emitted from the LED light source and generated hydrogen peroxide (¹O₂). In the cancer toxicity test, no cell toxicity was observed in HT-29 cancer cells and normal NIH-3T3 cells after 24-hour treatment with PaH alone. After 40 minutes of cultivation, the cell absorption of PaH reached its maximum, and PaH was localized in the mitochondria. The photodynamic treatment showed significant phototoxicity in HT-29 cells, especially in cells treated with PaH. As the concentration and light intensity increased, cell apoptosis increased significantly. Additional analysis showed that when the photodynamic treatment was performed at a concentration of 5 μM and an energy density of 10.8 J/cm², the initial and late cell apoptosis rates of HT-29 cells increased significantly to 21.54% and 5.39%, respectively. These results indicate that the natural photogenerating agent, phenylmethylsulfonate, has promising application potential in photodynamic therapy for squamous cell carcinoma.
