Nicotinamide adenine dinucleotide (abbreviated as NAD+ powder), also known as diphosphate pyridine nucleotide (abbreviated as DPN), or coenzyme I or coenzyme I. The oxidized form (NAD+) has a maximum ultraviolet absorption spectrum at 260nm. Through various deaminases, it accepts a hydrogen atom and an electron from the substrate to become a reduced form (NADH), which has a maximum absorption at 340nm.

As shown in the figure, a reversed-phase high-performance liquid chromatography (RP-HPLC) method for the detection of nicotinamide adenine dinucleotide (NAD+) was developed and validated. The method was then applied to the stability study of NAD+ in aqueous buffer solution and commercial dry test strips (Optium Plus™, Abbott Laboratories) for the determination of blood glucose.
NAD+ was separated from its decomposition products on an XTerra C18 column (150 mm × 4.6 mm, 5 µm) using a mobile phase consisting of a mixture of methanol and 20 mM potassium phosphate buffer pH 8.0 (5:95, v/v) at a flow rate of 1 mL/min with UV detection at 260 nm. The major decomposition products were identified as ADP-ribose and nicotinamide, and their contents depended on the characteristics of the buffer. The solid state stability of NAD formulated in Optium Plus™ test strips containing BES buffer at pH 7.0 was determined to be acceptable; >80% NAD+ remained after 3 months at 50°C.
