Lovastatin API CAS 75330-75-5 is a hexahydronaphthalene ester isolated from Aspergillus terrestris culture. It is an inactive lactone that becomes active only after hydrolysis. In the liver, it exhibits a strong competitive inhibitory effect on HMG-CoA reductase. HMG-CoA reductase is the rate-limiting enzyme in de novo cholesterol synthesis. Inhibition of this enzyme blocks the conversion of HMG-CoA to methylvalerate, significantly reducing cholesterol synthesis, leading to increased expression of hepatic LDL receptors and enhanced clearance of plasma LDL-C. Reduced cholesterol synthesis also reduces hepatic ApoB100 synthesis, thereby decreasing VLDL synthesis.
The structures of statins are very similar among naturally occurring statins. They share the same hydroxylhexahydronaphthalene polyketide moiety, with different side chains attached at C8 (R1) and C6 (R2) positions. Lovastatin has a methylbutyryl side chain at R1 and a 6-α-methyl side chain at R2; mevastatin lacks a 6-methyl group. Natural statins exist as lactones and require enzymatic hydrolysis to reach their acidic form in vivo to become active. While fully synthetic statins differ structurally from natural statins, they still possess an open-ring lactone moiety, sharing the common structure of all statins that competitively inhibits HMG-CoA reductase.

1.
