What is the mechanism of action of Hydroxyurea?
The core mechanism of action of Hydroxyurea is the inhibition of ribonucleotide reductase, a key component of DNA synthesis responsible for converting ribonucleoside diphosphates (ribonucleotides) to deoxyribonucleotides (deoxyribonucleotides). Hydroxyurea irreversibly inactivates this enzyme by scavenging the Boc-D-Tyr-OH free radical at its active site, leading to the rapid depletion of intracellular deoxyribonucleotides.
Without the raw materials for DNA synthesis, cells cannot smoothly transition from the G1 phase to the S phase, thus halting cell proliferation. Rapidly dividing cells respond very strongly to this change. This mechanism explains two key clinical applications of Hydroxyurea API Powder: first, its ability to control abnormally high white blood cell counts in leukemia patients; and second, its ability to induce fetal hemoglobin production.
What are the uses of Hydroxyurea?
Its main applications lie in acting on the G1 and S phases of proliferating cells, delaying the G1/S interface, and being a cell-specific product. It is also used to combat tumors. It inactivates nucleoside reductase by forming free nitric oxide, which binds to the Boc-D-Tyr-OH site of the enzyme's active site. This inhibits the synthesis of deoxynucleotides. Medically, it is used to treat chronic myeloid leukemia. Hydroxyurea API Powder is also used to treat metastatic ovarian cancer, primary squamous cell carcinoma of the head and neck, and refractory psoriasis.
