edoxaban is a small-molecule oral anticoagulant developed by Daisankyo Co., Ltd. in Japan, and it is an inhibitor of coagulation factor X(FXa). In the process of coagulation, activated coagulation factor X(FXa) activates coagulation plasminogen (FII) into thrombin (FIIa), which promotes the formation of fibrin, thus forming thrombus, so FXa has become the main target of developing a new generation of anticoagulant drugs. edoxaban powder is white to off-white, DMSO (slightly soluble, heated) and methanol (slightly soluble, heated).

Edoxaban powder has anticoagulant effect, which is mainly used for the prevention of stroke in patients with non-valvular atrial fibrillation and the prevention of venous thromboembolism after hip and knee replacement and hip fracture surgery. Edushaban is the fourth new oral anticoagulant approved by FDA after dabigatran, rivaroxaban and apixaban.
Preparation method of edoxaban
At room temperature, N-(5- chloropropidine -2- yl) -N'-[(1S,2R,4S)-4-(N,N- dimethyl formylamino) -2- (amino tert-butoxycarbonyl) cyclohexyl] oxamide (95.1g) was suspended in acetonitrile (190066ml). Triethylamine (155ml), 4,5,6,7- tetrahydro -5- methyl-thiazolo [5,4-c] pyridine -2- carboxylic acid hydrochloride (52.5g), 1- hydroxybenzotriazole (33.0g) and 1- ethyl-(3) were added to the reaction solution under ice cooling. Edoxaban powder was prepared by addingTriethylamine and water were added, stirred under ice cooling for 1 hour, and then crystals were filtered to obtain compound (1), namely 103.2g of etoxaban.
