Devimistat CAS 95809-78-2 is a first-in-class mitochondrial metabolism inhibitor. As an analogue of lipoic acid, it specifically targets the energy metabolism pathways of tumor cells and is currently mainly in the clinical research stage for the exploration of treatments for various cancers.

In vitro, CPI-613 exhibited selective toxicity against various tumor cell lines, including H460 human lung cancer cells and Saos-2 human sarcoma cells, with EC50 values ​​of 120 μM and 120 μM, respectively. CPI-613 interfered with mitochondrial metabolism in H460 cancer cells, including inhibiting PDH complex activity and cell membrane potential loss in a time- and dose-dependent manner. Furthermore, in both H460 human lung cancer cells and Saos-2 human sarcoma cells, CPI-613 (240 μM) induced apoptotic and non-apoptotic cell death.

CPI-613 represents a new direction in cancer treatment-targeting tumor energy metabolism, unlike traditional DNA-damaging chemotherapy products. Phase III clinical trials (such as those for pancreatic cancer) are currently underway, and the results will determine whether it can become a new clinical treatment option.
