It has the functions of an antioxidant and an effective scavenger of hydrogen peroxide radicals, protecting cell membrane lipids from the oxidation of these active radicals. It is generally used in biochemical research, analytical chemistry and synthetic bezoar. It can also be used as an antioxidant to protect membrane phospholipids from oxidation by hydrogen peroxide free radicals, and can protect neurons from oxidative damage even at nanomolar concentrations. It is also used for the determination of artificial bezoar bilirubin. Bilirubin has a variety of pharmacological effects and is the main raw material for making artificial bezoar. Pharmacological experiments have shown that it has a good inhibitory effect on W256 tumors, and its inactivation rate and inhibition index of Japanese encephalitis virus are 1 to 1.5 times higher than those of deoxycholic acid and bile acid; it is also an effective therapeutic drug for liver diseases. Without destroying liver tissue, it has the function of proliferating new cells and can treat serum hepatitis, cirrhosis and other diseases. In addition, bilirubin has sedative, anticonvulsant, antipyretic, and antihypertensive effects. It promotes the regeneration of red blood cells.
2. Improved calcium salt-chloroform extraction method Preparation of bilirubin calcium salt: Add sodium hydroxide to fresh pig bile to adjust pH 12, heat to boiling, add calcium chloride solution, heat to boiling again, cool, filter, and obtain bilirubin calcium salt. Pig bile [NaOH, CaCl2] → [pH 12, 100℃] Preparation of bilirubin calcium salt bilirubin finished product: Add 1:1 hydrochloric acid to bilirubin calcium salt to adjust pH 1-2, drain the acid water, add chloroform to extract, then evaporate the chloroform, add anhydrous ethanol to wash, and place the solution at about 5℃ to obtain the bilirubin finished product.
Alkaline hydrolysis: Measure 1000ml of fresh bile (filtered through double gauze), add a small amount of solid sodium bisulfite and 5ml of chloroform, and heat to 60-70℃. When cooled to 50℃ with constant stirring, add 0.2% sodium bisulfite solution, and add 2mol/L sodium hydroxide solution to adjust the pH to 11-12. Heat and boil for 3min with constant stirring, and cool to room temperature to obtain alkaline hydrolyzate. Fresh bile [NaHSO3, chloroform] → [60-70℃][0.2%NaHSO3] → [50℃][pH11-12] → [boiling 3min][room temperature] → alkaline hydrolyzate Chloroform extraction Add 0.1% sodium bisulfite solution and 30% chloroform to the alkaline hydrolyzate.
