Amlodipine powder is a long-acting calcium channel blocker that can be used to treat cardiovascular diseases such as angina pectoris, hypertension, and congestive heart failure. Amlodipine can reduce the influx of calcium ions, and its ability to inhibit calcium-induced aortic contraction is twice that of nifedipine. It can reduce myocardial contractility, slow heart rate, relax vascular smooth muscle, reduce peripheral resistance, and reduce cardiac load, thereby reducing myocardial oxygen consumption. Amlodipine can also dilate the epicardial and endocardial coronary arteries, increase coronary artery flow, improve blood supply or oxygen supply to ischemic areas, and relieve angina pectoris caused by coronary artery spasm.
Method 1: Ethyl chloroacetoacetate, N-benzylaminoethanol and sodium hydride react in tetrahydrofuran to obtain ethyl 4-(2-benzylaminoethoxy)acetoacetate. Then react with o-chlorobenzaldehyde, methyl 3-aminobutenoate and acetic acid in methanol, and the resulting product is hydrogenated and reduced to obtain amlodipine. It is salified with maleic acid to obtain its maleate.
Method 2: Ethyl chloroacetoacetate and azidoethanol are condensed under the action of sodium hydride to obtain ethyl 4-(2-azidoethoxy)acetoacetate, which is then cyclized with o-chlorobenzaldehyde and methyl 3-aminobutenoate, and finally the azido group is reduced to obtain amlodipine.HPLC spectrum is shown in the figure.
