In more details, the engineered yeast was transferred to an SD-URA medium using plate streaking, and it was incubated at 30 °C for 56 h. A single colony was then selected and cultured in a liquid medium, followed by overnight incubation at 30 °C and 220 rpm to generate the seed liquid. Subsequently, the seed solution was inoculated into a liquid medium containing trans-2-decenoic acid (OD = 0.4) and cultured for 24 h. The supernatant was collected, and the pH was adjusted to 11–12 using 20 % NaOH. The mixture was left to separate for 1–4 h. The supernatant was then filtered through 0.22 μm and 300-Da ultrafiltration membranes, and the filtrate was collected. Activated carbon was added for decolorization for 1 h. The solution was transferred to the plate and frame filtration device to collect the filtrate. The pH was adjusted to 2.0–3.0 with 1 mol/L H2SO4, cooled to 20 °C, and crystallized for 5 h to allow the effective ingredients to crystallize and precipitate. The solution was then transferred again to the plate and frame filtration device to collect the filter cake. It was dissolved in 95 % ethanol, diluted with three times the volume of deionized water, and the filter cake was collected again after crystallization. The filter cake was placed in a vacuum dryer, and the temperature was raised to 40–50 °C. Drying for 1–4 h resulted in the production of 10-HDA (D).
